How does Nyclobate 0.05% Cream work?

It acts by blocking the release of certain chemical mediators (prostaglandins, leukotrienes) in the body that cause pain, inflammation, and swelling. As a result, it effectively reduces the skin's redness, itchiness, inflammation, swelling, and irritation associated with certain skin problems.

Side Effects of Nyclobate 0.05% Cream

Quick Suggestions:

• It is for external use only. Wash your hands before and after application. Apply a thin layer of Nyclobate 0.05% Cream on the affected area. Do not cover the affected site after applying Nyclobate 0.05% Cream.
• Do not cover the area being treated with airtight dressings such as bandages unless directed by a doctor, as this may increase the risk of side effects.
• If you think the area of skin you are treating has become infected you should stop using Clobetasol and consult your doctor.
• Consult your doctor if your skin condition has not improved after four weeks of treatment. Do not use it for more than 4 consecutive weeks at a time.

Clobetasol propionate is indicated for the treatment of corticosteroid-responsive dermatoses, including:

• Psoriasis (scalp and plaque-type psoriasis)
• Vitiligo
• Eczema
• Alopecia areata
• Herpes labialis
• Lichen sclerosus
• Mycosis fungoides

Pharmacology

Clobetasol Propionate is a highly potent topical corticosteroid with anti-inflammatory, antipruritic, and vasoconstrictive properties. It acts by reducing mast cells, inhibiting eosinophil activation, reducing cytokine production, and inhibiting arachidonic acid metabolism, thus controlling allergic responses.

Dosage & Administration of Nyclobate 0.05% Cream

Adult Dosage

• Corticosteroid-Responsive Dermatoses (Cream): Apply a thin layer to the affected areas every 12 hours and rub in gently. Do not exceed 50 g per week.
• Scalp Psoriasis (Shampoo): Apply a thin film to the dry scalp once daily, leave it in place for 15 minutes, then add water, lather, and rinse thoroughly.
• Moderate to Severe Plaque-type Psoriasis (Cream/Lotion): Apply to the affected area of the scalp every 12 hours for up to 2 weeks if the area is less than 10% of the body surface area. Do not exceed 50 g per week.

Pediatric Dosage

• <12 years: Safety and efficacy not established.
• >12 years:
  • Corticosteroid-Responsive Dermatoses (Cream): Apply a thin layer to the affected areas every 12 hours and rub in gently. Do not exceed 50 g per week.
  • Scalp Psoriasis (Shampoo): Apply a thin film to the dry scalp once daily, leave it in place for 15 minutes, then lather and rinse thoroughly.
  • Moderate to Severe Plaque-type Psoriasis (Cream/Lotion): Apply to the affected area of the scalp every 12 hours for up to 2 weeks if the area is less than 10% of the body surface area. Do not exceed 50 g per week.

Apply creams for moist surfaces and ointments for dry, scaly lesions.

Contraindications

• Rosacea, acne vulgaris, perioral dermatitis
• Primary cutaneous viral infections (e.g., herpes simplex, chickenpox)
• Fungal and bacterial infections, perianal and genital pruritus
• Children under 1 year of age

Pregnancy and Lactation Safety for Clobetasol

Pregnancy

• Safety data on the use of clobetasol in pregnant women is not available.
• Published reports indicate a significantly increased risk of low birth weight in babies when more than 300 grams of potent or very potent topical corticosteroids, such as clobetasol, are used during pregnancy.
• Pregnant women should be advised of the potential risk to the fetus.
• To minimize risks, clobetasol should be used on the smallest area of the skin and for the shortest duration possible.
• Animal studies have shown increased risks of malformations, such as cleft palate and skeletal abnormalities, after subcutaneous (SC) administration of clobetasol to pregnant mice and rabbits.

Lactation

• There is no information available regarding the presence of clobetasol in breast milk, its effects on breastfed infants, or its impact on milk production.
• Systemically administered corticosteroids can appear in human milk and may cause:
  • Growth suppression
  • Interference with endogenous corticosteroid production
  • Other potential adverse effects
• It is unknown whether topical use of clobetasol can result in sufficient systemic absorption to produce detectable quantities in breast milk.
• To minimize exposure to the breastfed infant, clobetasol should be used on the smallest area of skin and for the shortest duration possible.
• Breastfeeding women should avoid applying clobetasol directly to the nipple and areola to prevent direct exposure to the infant.

Precautions & Warnings for Clobetasol Propionate

• Avoid long-term use, especially in children, to prevent adrenal suppression.
• Use with caution on the face and around the eyes due to the risk of atrophy, glaucoma, or cataracts.
• Careful supervision is required in treating psoriasis due to risks like rebound psoriasis and toxicity.
